Patent Title: Fish-in-net immobilized enzyme based on molecular imprinting
Publication date: 2010
Publication number: CN101434947
Abstract: The invention discloses a fish-in-net immobilized enzyme based on molecular engram, which belongs to the fields of inorganic chemistry, physical chemistry and biochemistry. The immobilized enzyme takes soluble salts of silicon or/and metal oxide as raw material, forms colloidal sol by taking a surface active agent as a modeling agent, and combines the enzyme and the colloidal sol into an inorganic bioreactor under the protection of engram molecules. The fish-in-net immobilized enzyme takes a shell of inorganic material, a catalytic active centre of enzyme, enzyme system and enzyme reagent that are related to carbohydrate, and the engram molecules of small carbohydrate molecules in enzyme substrates and/or products. As traditional immobilized enzymes have bad enzyme activity preservation, the method adds the enzyme engram molecules into the immobilization of enzyme, enzyme system and enzyme reagent, and the engram molecules realize stable catalytic system in the immobilization process from two aspects: (1) similar to the action of the engram molecules, the engram molecules protect the conformation of enzyme active centre and catalytic activity; and (2) hydration sites on the enzyme surface are substituted, thus lowering the enzyme modification action that is caused by organic medium in the immobilization, and immensely preserving the enzyme activity.
